White bands on film??
« on: November 03, 2011, 12:30:10 AM »
Hi all,

Lately I got a question during a presentation around white bands on film when working with Western blotting. Why does this white band occur?

Most likely excessive signal has been generated. So how to avoid it? Reduce antibody or protein concentration as excessive quantities can cause extremely high levels of localized signal (usually at a single band) leasing to rapid complete consumption of substrate. At this point-as no light can be generated after the completion of this reaction, white bands appear when exposed.

Hope this can be to some help!
